EANM guidance document: dosimetry for first-in-human studies and early phase clinical trials

The numbers of diagnostic and therapeutic nuclear medicine agents under investigation are rapidly increasing. Both novel emitters and novel carrier molecules require careful selection of measurement procedures. This document provides guidance relevant to dosimetry for first-in human and early phase clinical trials of such novel agents. The guideline includes a short introduction to different emitters and carrier molecules, followed by recommendations on the methods for activity measurement, pharmacokinetic analyses, as well as absorbed dose calculations and uncertainty analyses. The optimal use of preclinical information and studies involving diagnostic analogues is discussed. Good practice reporting is emphasised, and relevant dosimetry parameters and method descriptions to be included are listed. Three examples of first-in-human dosimetry studies, both for diagnostic tracers and radionuclide therapies, are given.

Validation of the dew point chilled mirror method for the measurement of water activity on solid pharmaceutical products, testing the effect of different temperatures, sample preparations, and environment exposure times

Abstract Objective The objective of this investigation is to validate the dew point chilled mirror method for quantifying water activity in tablets and capsules, evaluating the effects of different temperatures, sample preparations, and local environmental exposure times. Key findings The validation of an acute and precise water activity measurement method has become an important goal in the pharmaceutical industry, because it may help to predict the microbiological bioburden on solid products, since most xerophilic and osmophilic microorganisms are unable to grow at water activity levels below 0.60, to safely reduce the frequency of routine microbiological analysis using conventional methods. For all the solid samples tested, a suitability of method was carried out, considering the sample preparation, environmental exposure times, and different measuring temperatures. Following USP guidelines, essential parameters such as precision (SD < 0.5), accuracy (% recovery in the 95%–105% range), linearity (R2 > 0.99), ruggedness (ANOVA, P < .05), robustness, operative range (aw 0.17–1), limits of detection (aw = 0.17), and limit of quantification (aw = 0.25) were met by the dew point methodology. Conclusion The dew point chilled mirror method was proven to yield accurate, precise, and robust data, making it an outstanding methodology to be implemented in the pharmaceutical industry for measuring the water activity status in tablets and capsules as a direct assessment of the microbial burden.

Phytofabrication of silver nanoparticles using callus extracts of natural tetraploid Trifolium pratense L. and its bioactivities

One of the main subjects of plant biotechnology is plant tissue culture and in recent years is considered a possible approach model for green and eco-friendly biosynthesis of nanoparticles. This study aimed to present calli produced from the natural tetraploid Trifolium pratense L. containing high amounts of phenolic compounds and glycosidic bioactive macromolecules and the biosynthesis of silver nanoparticles from calli. Combinatorial optimization of silver nanoparticles was achieved for the first time in this study, thanks to the stabilizing and reducing properties of hypocotyl, apical meristem, and epicotyl derived callus extracts of the natural tetraploid T. pratense L. biosynthesized nanoparticles from three different callus extracts. Callus extracts were used to create different experiments with AgNO3 at various concentrations (0.16, 0.5, 0.84, 1.18, 1.52 and 1.96 mg L-1), different temperatures (40, 50, 60, 70, 80, 90, 100°C), and different pH levels (5, 7, 10) to carry out the biosynthesis of AgNPs. Biologically synthesized AgNPs were easily monitored by color change in ultraviolet and UV-Vis spectroscopy proved to be a fast and simple method. Also, TEM, XRD, and FTIR analyses were done to characterize and confirm the formation of crystalline nanoparticles. It was determined that antibacterial activity inhibition was achieved by using the Agar-well diffusion method for antibacterial activity measurements on Gram-positive Staphylococcus aureus ATCC 25923 and Gram-negative Escherichia coli CECT 4972 bacteria. Biosynthesized AgNPs were observed in the wavelength range of 400-500 nm in the UV-VIS spectrum. TEM analysis demonstrated the size and shape of biosynthesized silver nanoparticles under different conditions. It was observed that the smaller silver nanoparticles were spherical and the larger silver nanoparticles were triangular, elliptical, and spherical shape. The XRD analysis proved the presence of Ag0 in nanoparticles and showed crystal structure for silver nanoparticles. By FTIR analysis, O-H hydroxyl groups of functional groups on the AgNP surface, H-linked OH stretching, C-H stretching, -CH stretching of -CH2 and -CH3 functional groups, C-N and carboxylate, aliphatic phosphate and primary amine stretching were expressed. Biosynthesized silver nanoparticles showed antibacterial activity against Gram-positive S. aureus ATCC 25923 bacteria, AgNP hypocotyl (1.7mm), AgNP-epicotyl (1.1mm) against Gram-negative E. coli CECT 4972 bacteria. Among the hypocotyl, apical meristem, and epicotyl callus cultures, the highest antioxidant activity was observed in the AgNPs obtained from hypocotyl-concentration experiments, with a DPPH radical activity of 52% and an ABTS radical activity of 68%. In conclusion, these findings underscore the potential of biotechnological strategies in green nanotechnology, which can be offered for developing metal nanoparticles with potential biomedicine and biotechnology applications.

Physical activity of children and adolescents who use a wheelchair: a systematic review

Physical activity has numerous health benefits for people with physical disabilities. Nevertheless, activity levels are often below recommended levels. To promote physical activity among children and adolescents who use a wheelchair as their primary source of mobility, this systematic review explores the physical activity patterns of this group. A systematic search of PubMed, Sports Medicine & Education Index, Web of Science, and SPORTDiscus was performed, included articles were synthesized in terms of duration, intensity, and settings in which physical activity occurred, as well as the physical activity measurement methods. Nine articles were included. The mean overall physical activity level across the included studies was 98 minutes per day (range: 78–115 minutes per day). Two articles analysed the duration of physical activity at different intensities (very light physical activity, light physical activity (LPA), moderate to vigorous physical activity (MVPA) and intensities near to maximum). Within the included articles, both subjective and objective measurement methods were used. Due to the small number of articles, combined with small sample sizes, there is not enough evidence to answer the research questions sufficiently. Nevertheless, the review provides an overview of actual research and clearly shows that the physical activity values are insufficiently researched. There is a need for further research on the scope, types and settings of physical activity in the target group.

A quantitative assay system for protein C activity, the regulator of blood coagulation, based on a chromogenic method mimicking the blood coagulation cascade

Background and aimsProtein C is a plasma protein, and its active form regulates blood coagulation. The recommended unit of protein C activity is IU/mL; however, some laboratories use percentage. Some deficiencies cannot be detected owing to measurement principles. This study sought to quantify protein C activity levels and overcome the limitations of the current measurements. Materials and methodsOur protein C activity measurement method mimicked the blood coagulation cascade and used a thrombin-specific chromogenic reagent. The control was prepared by adding protein C to the protein C deficient plasma. The calibration curve was plotted as the increase in the absorbance per minute and the concentration of protein C in the control. Statistical tests were performed to compare our method with the current chromogenic method. ResultsA calibration curve was constructed (y = −0.0132x + 0.14, R2 = 0.9987, n = 10). The statistical results of our method suggested non-inferiority when compared to the current chromogenic method (α = 0.05). ConclusionThe quantitative measurement was performed using plasma samples. Our method provides the possibility of expressing protein C activity quantitatively and detecting deficiencies that cannot be detected using the current chromogenic method.

Modeling categorical time-to-event data: The example of social interaction dynamics captured with event-contingent experience sampling methods.

The depth of information collected in participants' daily lives with active (e.g., experience sampling surveys) and passive (e.g., smartphone sensors) ambulatory measurement methods is immense. When measuring participants' behaviors in daily life, the timing of particular events-such as social interactions-is often recorded. These data facilitate the investigation of new types of research questions about the timing of those events, including whether individuals' affective state is associated with the rate of social interactions (binary event occurrence) and what types of social interactions are likely to occur (multicategory event occurrences, e.g., interactions with friends or family). Although survival analysis methods have been used to analyze time-to-event data in longitudinal settings for several decades, these methods have not yet been incorporated into ambulatory assessment research. This article illustrates how multilevel and multistate survival analysis methods can be used to model the social interaction dynamics captured in intensive longitudinal data, specifically when individuals exhibit particular categories of behavior. We provide an introduction to these models and a tutorial on how the timing and type of social interactions can be modeled using the R statistical programming language. Using event-contingent reports (N = 150, Nevents = 64,112) obtained in an ambulatory study of interpersonal interactions, we further exemplify an empirical application case. In sum, this article demonstrates how survival models can advance the understanding of (social interaction) dynamics that unfold in daily life. Colorimetric Assay for Protein Redox Activity – a Complement to Electrochemistry

Oxidoreductases are vital biocatalytic tools for driving redox conversions that are challenging using conventional catalysts. Monooxygenases, an important class of these enzymes, have been the subject of enzyme engineering due to their high activity and versatile substrate scope. Reactions performed by these biocatalysts have long been monitored by a colorimetric method involving the coupling of a dye precursor to naphthalene hydroxylation products generated by the enzyme. Despite the popularity of this method, we found the dye product to be unstable, preventing quantitative readout. By incorporating an extraction step to solubilize the dye produced, we have improved this assay to the point where quantitation of enzyme activity is possible. Further, by incorporating spectral deconvolution, we have, for the first time, enabled independent quantification of the two possible regioisomeric products: 1-naphthol and 2-naphthol. Previously, such analysis was only possible with chromatographic separation, increasing the cost and complexity of analysis. The efficacy of our improved workflow was evaluated by monitoring the activity of a toluene-4-monooxygenase enzyme from Pseudomonas mendocina KR-1. Our colorimetric regioisomer quantification was found to be consistent with chromatographic analysis by HPLC. The development and validation of a quantitative colorimetric assay for monooxygenase activity that enables regioisomeric distinction and quantification represents a significant advance in analytical methods to monitor enzyme activity. By maintaining facile, low-cost, high-throughput readout while incorporating quantification, this assay represents an important alternative to more expensive chromatographic quantification techniques. For the electrochemistry community, the use of monooxygenases or other redox capable enzymes for biocatalysis must be complemented by a toolkit of methods for accurate enzyme activity measurement. Our approach demonstrates that accurate, mixture analysis is possible in a higher throughput using the improved colorimetric method. The relevant assay reaction, the hydroxylation of naphthalene, may be readily incorporated in the future for measurement schemes with other enzymes than the tested toluene-4-monooxygenase, thus enabling opportunities to tune these biocatalysts towards desired activities.

Study of radium content and radon exhalation rates in raw building materials used in southern India

As a potential source of indoor radiation arising from radon, a range of building materials (white cement, gray cement, gravel, gypsum, granite, marble, vitrified tile, and brick) were studied for the specific activity of radium and radon gas exhalation rates. The activity of radium was determined using scintillation gamma-ray spectrometry. Exhalation rates were assessed using passive and active measurement methods. Radon mass exhalation rates from powdered building materials and radon surface exhalation rates from slab-type building materials were measured. Activity concentrations of 226Ra in powder and slab-type building materials were found in the range of (1.12 – 14.98) Bqkg−1 and (17 – 308) Bqkg−1, respectively. 222Rn mass exhalation rates (mBqkg−1h−1) from the building materials were in the range of 15.0 ± 0.8 (gray cement) to 1.1 ± 0.1 (gravel). Surface exhalation rates (mBqm−2h−1) for slab-type materials have a range of 317 ± 9 (brick) to 8 ± 2 (marble). Strong positive correlations were found between the specific activity of radium and radon mass exhalation rates (r = 0.9408) and radon surface exhalation rates (r = 0.7167) for building materials investigated. Obtained data indicates that the building materials used influence indoor radon concentration in dwellings.

Quantitative assessment of heavy-duty anticorrosion coating thickness via near-infrared measurements

The application of heavy-duty anticorrosion coatings is a typical method for preventing corrosion in steel bridges. However, thus far, no efficient and quantitative inspection technique has been established that can remotely and non-destructively evaluate the remaining thickness of the top coat in anticorrosion coatings. This study proposes a new inspection method called the "active self-reference lock-in near-infrared measurement method", which can accurately and quantitatively assess the remaining thickness of the top coat, irrespective of the lighting conditions. Experimental studies conducted on specimens and field studies carried out on on-site bridge measurements have confirmed the efficacy of the proposed method.

Development of brake activity measurement method for heavy-duty vehicles

ABSTRACT Tailpipe PM (particulate matter) emissions have been reduced due to decades of tightening regulations, however non-tailpipe PM emissions are not regulated and are expected to become a significant source of traffic-related PM emissions. Previous studies have focused on emission measurement from laboratory and track tests. Their findings suggest brake wear PM emission rates are dependent on brake activity. Therefore, it is important to characterize brake emissions by first understanding the real-world brake activity from many different vehicle vocations and driving conditions. The goal of the current study is to establish a test method and analysis for brake activity measurements of heavy-duty vehicles. In this study, brake fluid pressure and brake pad temperature were measured for a heavy-duty vehicle during chassis and on-road driving tests. The chassis tests consisted of the Central Business District (CBD) cycle representative of a repetitive stop-and-go driving pattern of a bus, and the Urban Dynamometer Driving Schedule (UDDS) cycle representative of urban driving conditions of heavy-duty vehicles. The on-road tests consisted of a local Riverside City route focused on urban roads at low vehicle speeds with frequent braking, while the second route from Riverside City to Victorville focused on highway driving and downhill braking. The brake pad temperature of the triplicate CBD cycle gradually increased linearly with a slope of 2.3°C/min and the temperature per kinetic energy lost during braking increased by 2.3 × 10−5°C/J for the CBD cycle. The UDDS cycles had the largest kinetic energy loss between 3.2 × 103 to 3.0 × 105 J in the histogram. The local Riverside city route brake temperature increased by 2.0°C/min. The kinetic energy loss for the on-road tests were one order of magnitude larger than that of the dynamometer tests due to brake events occurring under higher speeds. Implications: The non-tailpipe source contributions to traffic related particulate matter (PM) emissions have surpassed that of tailpipe emissions. The results of this work provide a measurement method to obtain brake activity information for a heavy-duty vehicle, which is critical estimating emission inventory accurately.

In this study, a subjective experiment was conducted using both a virtual environment technology and an electrodermal activity measurement method. This study is a part of research aimed at obtaining knowledge about window width that does not cause mental strain. The study results revealed that 1) These results mainly indicate that the narrower the window width, the higher the mental strain on the person staying in a room. 2) The results also suggested that mental strain varied depending on the type of avatar. These findings might be valuable when creating a window arrangement that results in less mental strain.

The In Vitro Impact of Isoxazole Derivatives on Pathogenic Biofilm and Cytotoxicity of Fibroblast Cell Line.

The microbial, biofilm-based infections of chronic wounds are one of the major challenges of contemporary medicine. The use of topically administered antiseptic agents is essential to treat wound-infecting microorganisms. Due to observed microbial tolerance/resistance against specific clinically-used antiseptics, the search for new, efficient agents is of pivotal meaning. Therefore, in this work, 15 isoxazole derivatives were scrutinized against leading biofilm wound pathogens Staphylococcus aureus and Pseudomonas aeruginosa, and against Candida albicans fungus. For this purpose, the minimal inhibitory concentration, biofilm reduction in microtitrate plates, modified disk diffusion methods and antibiofilm dressing activity measurement methods were applied. Moreover, the cytotoxicity and cytocompatibility of derivatives was tested toward wound bed-forming cells, referred to as fibroblasts, using normative methods. Obtained results revealed that all isoxazole derivatives displayed antimicrobial activity and low cytotoxic effect, but antimicrobial activity of two derivatives, 2-(cyclohexylamino)-1-(5-nitrothiophen-2-yl)-2-oxoethyl 5-amino-3-methyl-1,2-oxazole-4-carboxylate (PUB9) and 2-(benzylamino)-1-(5-nitrothiophen-2-yl)-2-oxoethyl 5-amino-3-methyl-1,2-oxazole-4-carboxylate (PUB10), was noticeably higher compared to the other compounds analyzed, especially PUB9 with regard to Staphylococcus aureus, with a minimal inhibitory concentration more than x1000 lower compared to the remaining derivatives. The PUB9 and PUB10 derivatives were able to reduce more than 90% of biofilm-forming cells, regardless of the species, displaying at the same time none (PUB9) or moderate (PUB10) cytotoxicity against fibroblasts and high (PUB9) or moderate (PUB10) cytocompatibility against these wound cells. Therefore, taking into consideration the clinical demand for new antiseptic agents for non-healing wound treatment, PUB9 seems to be a promising candidate to be further tested in advanced animal models and later, if satisfactory results are obtained, in the clinical setting.

Evaluation of a simple activity measurement method in rats.

[Purpose] Behavioral analysis is widely used in animal research. However, such analysis requires specialized equipment and can be difficult to perform. Therefore, this study aimed to explore and validate a simple behavioral analysis method. [Participants and Methods] For behavioral assessments, Wistar rats were placed in a rearing cage and videotaped from two directions: overhead and side view. The filmed videos were analyzed using ImageJ software to calculate the distance traveled and activity and inactivity times of the rats. Intraclass correlation coefficients 1 and 2 were calculated to examine the reliability of the behavioral analysis method. [Results] Intraclass correlation coefficients 1 and 2 for distance traveled and activity and inactivity times determined using the behavioral analysis method showed high reliability. [Conclusion] The behavioral analysis method validated in this study used inexpensive and easily accessible equipment and devices. The results show high correlation coefficients for the measurement of distance traveled and activity time performed by experimental animals, demonstrating the reliability of this simple method.

The antibiofilm activity of selected substances used in oral health prophylaxis

Oral health is a window to a patient’s general well-being. Balance in oral microbiome functions is crucial for health maintenance. A state of oral dysbiosis may lead to a variety of local and systemic pathological conditions. The presence of dental plaque is related to the majority of oral infections. Proper oral hygiene is crucial and the most economic practice contributing to oral health prophylaxis. Aside from prophylactic treatments provided by dental practitioners, mouth rinses, containing antimicrobial agents, are one of the possible tools used for oral care. Our study was to determine whether available mouth rinses and selected products dedicated for professional use are efficient to eradicate biofilm formed by reference and clinical strains of Streptococcus mutans, Streptococcus sanguinis, Streptococcus oralis, Streptococcus mitis, Staphylococcus aureus, Enterococcus faecalis, Lactobacillus rhamnosus and Candida albicans on the surface of hydroxyapatite – major mineral component of a tooth. Therefore, such antimicrobials as chlorhexidine, cetylpyridine chloride, polyhexanide, silver nanoparticles, sulphonated phenolics, and natural antiplaque essential oils and coconut oil were analyzed. Applied experimental settings in in vitro models were designed to reflect accurately the recommended use of the tested substances, therefore four types of eradication procedure were conducted. Sialorrhea simulation was also performed to evaluate antibiofilm potential of diluted mouth rinses. Biofilm was investigated with quantitative method where absorbance values were measured. Statistical differences were assessed using the Kruskal–Wallis test with a post-hoc Dunnett’s analysis. Results have shown that biofilms displayed a diversified sensitivity to the tested antimicrobials. The highest antibiofilm activity was detected for cetylpyridine chloride while the lowest for chlorhexidine. However the differences in E. faecalis biofilm reduction observed after the use of these two compounds were not statistically significant (p > 0.05), whereas all observed differences in S. aureus survival after exposure to the examined antimicrobial agents were statistically significant (p < 0.5). The PHMB, both in standard and in sialorrhea simulated conditions had the highest potential against streptococci. The coconut oil reduced C. albicans fungus biofilm by 65.48% but low eradication level was observed in case of bacterial biofilms. The dehydrating mechanism of action of sulfonated phenolics turned out to be ineffective against streptococcal biofilm which in turn was effectively eradicated by silver nanoparticles. The implementation of Antibiofilm Dressing's Activity Measurement method allowed to observe strain-related differences in terms of antimicrobial sensitivity. The obtained results may be introduced in everyday out-patient dental plaque prophylaxis as well as clinical environment.

Integrative interaction of emotional speech in audio-visual modality.

Emotional clues are always expressed in many ways in our daily life, and the emotional information we receive is often represented by multiple modalities. Successful social interactions require a combination of multisensory cues to accurately determine the emotion of others. The integration mechanism of multimodal emotional information has been widely investigated. Different brain activity measurement methods were used to determine the location of brain regions involved in the audio-visual integration of emotional information, mainly in the bilateral superior temporal regions. However, the methods adopted in these studies are relatively simple, and the materials of the study rarely contain speech information. The integration mechanism of emotional speech in the human brain still needs further examinations. In this paper, a functional magnetic resonance imaging (fMRI) study was conducted using event-related design to explore the audio-visual integration mechanism of emotional speech in the human brain by using dynamic facial expressions and emotional speech to express emotions of different valences. Representational similarity analysis (RSA) based on regions of interest (ROIs), whole brain searchlight analysis, modality conjunction analysis and supra-additive analysis were used to analyze and verify the role of relevant brain regions. Meanwhile, a weighted RSA method was used to evaluate the contributions of each candidate model in the best fitted model of ROIs. The results showed that only the left insula was detected by all methods, suggesting that the left insula played an important role in the audio-visual integration of emotional speech. Whole brain searchlight analysis, modality conjunction analysis and supra-additive analysis together revealed that the bilateral middle temporal gyrus (MTG), right inferior parietal lobule and bilateral precuneus might be involved in the audio-visual integration of emotional speech from other aspects.

Development of a New Colorimetric, Kinetic and Automated Ceruloplasmin Ferroxidase Activity Measurement Method.

Ceruloplasmin plays an important role in the regulation of iron metabolism. Ceruloplasmin is an acute-phase protein known to have many metabolic effects. Its activity increases during infection, inflammation, and compensation of oxidation. In the current study, our aim is to develop a new method for the measurement of ferroxidase activity without requiring any chromogen. Venous blood samples were collected into serum separator tubes. Ferric iron ions formed by the enzyme ferroxidase were measured, both manually and fully automatically, at the 415 nm wavelength without using chromogen. These results were compared to conventional ferroxidase measurement methods and to the immunoturbidimetric ceruloplasmin measurement method. The detection limit of the new assay was 14.8 U/L. The upper limit of the linearity was 1380 U/L. Precision values were calculated for high, medium, and low levels of ferroxidase activity in serum pool. The coefficient of variation was &lt;5% for each level. In the present method, chromogens are not used. With its considerably low cost and short reaction time, this method is able to provide fast results, can be performed easily, and makes accurate measurements.

Design of laser scanning binocular stereo vision imaging system and target measurement

Binocular stereo vision is an important scheme of road environment perception for intelligent driving, because it can provide image information and spatial three-dimensional position information. However, binocular stereo matching is often disturbed by the environment, and noise, shadow, light transformation, and so on, will all cause mismatching, therefore it is impossible to carry out accurate 3D reconstruction. In this paper, an active imaging perception and target measurement method combining laser scanning and binocular vision is proposed. The binocular vision structure is used to simulate the human eye's observation of the environment in front of the vehicle. At the same time, through the laser lines scanned to the target, the strong characteristics of binocular matching are provided to enhance the anti-interference of binocular matching, and calculate the three-dimensional point cloud data of the front object according to the binocular stereo vision theory. Firstly, this paper studies the influence of the parameters changing of the system on the accuracy of object depth value in indoor environment, and then selects the most suitable system parameters to carry out three-dimensional reconstruction experiment on the actual target object in indoor environment. Finally, the scene experiment in outdoor environment is carried out. In the outdoor environment, the red spectrum laser equipped in the system can still effectively reconstruct the target surface radian, but there are some limitations due to the limitation of outdoor light intensity. This paper preliminarily summarizes the current structural design and experiment, and further discusses the follow-up research and improvement ideas. Research and experiments show that the laser scanning binocular stereo vision imaging system proposed in this paper has the function of forward target ranging, especially under some bad imaging environment, this method can also effectively match the binocular images, and present the three-dimensional information of the internal details of the target. Under the indoor experimental environment, the minimum relative error is 1.42 % for the target with a distance of about 1.5 m, and in the evening outdoor environment, the minimum the minimum relative error is 0.87 % for the target with a distance of about 3 m. Due to the advantages of laser line intensity feature matching, it can accelerate the speed of binocular matching, and has potential application prospects in autonomous system.
